The neuronal and extraneuronal origins of plasma 3-methoxy-4-hydroxyphenylglycol in rats.

摘要: 3-Methoxy-4-hydroxyphenylglycol (MHPG) is formed by the sequential actions of monoamine oxidase (MAO) and catechol-O-methyltransferase on norepinephrine within extraneuronal tissues or O-methylation 3,4-dihydroxyphenylglycol (DHPG) produced intraneuronally from norepinephrine. This study examined contributions neuronal metabolism to formation MHPG in rats. Spillover into plasma (605 +/- 28 pmol/kg per min) was higher than spillover DHPG (463 15 min), (165 9 normetanephrine (56 4 min). Comparison with increment after MAO inhibition (81 combined derived circulating (21 indicated that only 17% deamination norepinephrine; remaining 83% neurons. The production infused 30% entry bloodstream 53% (83-30) before its bloodstream. Metabolism made a minor (1-3%) contribution concentrations MHPG, but much larger (46%) normetanephrine. results provide comprehensive examination pathways. Since principal metabolite excreted rat, findings also show total body turnover dependent mainly transmitter; at rest pathways contribute as little 15% turnover.
