Involvement of carbohydrate moieties of the toad egg vitelline coat in binding with fertilizing sperm

摘要: Vitelline coats (VC) were isolated from the eggs of Bufo japonicus, and added with sperm in reconstituted salt solution, which mimics physiological role jelly envelopes, to determine rates binding per unit area (0.2 mm2) VC. The rate VC uterine was high, but low coelomic activated 1/20 De Boer's solution (DB) moderately DB. increased when treated extracts pars recta portion oviduct. that bound not acrosome-reacted their required both a salinity, assuring motility sperm, sufficiently high levels Ca2+ Mg2+. reduced by either coexisting solubilized materials, periodate-oxidation or pretreatment Fab fragments anti-VC antibodies, reacted mostly carbohydrate residues glycoproteins. Sperm-VC assays combination gel-filtrated components revealed fractions containing 36–39 kDa most effective inhibiting neutralizing antibody induced inhibition binding. These results indicate moieties glycoproteins VC, exposed as result hydrolysis oviducal protease, are involved fertilizing sperm.
