摘要: The cloned mammalian cell entry gene mce1a from Mycobacterium tuberculosis confers to non-pathogenic Escherichia coli the ability invade and survive inside macrophages HeLa cells. aim of this work was search for characterize homologs four M. operons (mce1, mce2, mce3 mce4) in mycobacteria other than (MOTT). dot-blot polymerase chain reaction (PCR) experiments performed on 24 clinical isolates representing 20 different mycobacterial species indicated that mce were widely distributed throughout genus Mycobacterium. BLAST results showed presence mce1, mce2 mce4 bovis, avium smegmatis. A homologous region operon also found DNA protein alignments done compare deduced avium, smegmatis homologs. proteins bovis had 99.6–100% homology with respective (MTmce). similarity between individual ranged 56.2 85.5%. alignment MTmce 58.5% 68.5%. Primer sets designed mce4a amplification 379-bp fragments. Amplification successful 14 strains 11 species. PCR fragments sequenced 10 eight Alignment products are highly conserved In conclusions, generally organized same manner. phylogenetic tree comparing mce1 closely related diverged operons. wide distribution pathogenic implicates these putative virulence genes is not an indicator pathogenicity bacilli. Instead, factors might be determined by their expression.
