The Influence of Sample Mass and Flow Velocity on Performance in Preparative Column Liquid Chromatography

摘要: Abstract The H/u curves of two HPLC columns 10 mm i.d., filled with silica a particle diameter 5 and 25–40 μm, respectively, have been determined at various amounts injected sample. Indications mass overload, Le, peak broadening, occur sample load μg/g for the column μm but not until 100 coarse stationary phase. Although H values increase markedly higher mass, slopes remain almost constant on both up to samples 1 mg/g. This means relative plate height increasing flow velocity becomes nearly negligible under conditions moderate overload. However, by or mg/g, show dramatic slope: high overload mobile phase rate cannot be increased without remarkable decrease in performance.
