Amniotic membrane and adipose-derived stem cell co-culture system enhances bone regeneration in a rat periodontal defect model

摘要: Background/Purpose Periodontal disease is a chronic inflammatory process that potentially leads to alveolar bone destruction and tooth loss. Tissue engineering combined with stem cell therapy potential effective treatment for periodontal Amniotic membrane (AM) scaffold enriched multiple growth factors. It has the effects of anti-inflammation, antiangiogenesis, immunosuppression. Herein, we used adipose-derived cells (ADSCs) an AM co-cultured system study regeneration in rat defect model in vivo . Methods Human ADSCs were isolated from infrapatellar fat pad, characterized by flow cytometry, reverse transcription-polymerase chain reaction, multipotent differentiation assays. The co-culture was applied two-wall osseous model, computed tomography measure effect. Results pad showed spindle-like morphology. Flow cytometry results demonstrated expressed high level CD90 CD105, but not CD31, CD34, CD45. strongly stemness genes, including SOX2 , OCT4 NANOG KLF4 on different passages. Furthermore, able differentiate into osteogenic, chondrogenic, adipogenic cells. In significantly increased regeneration. Conclusion This provides basis using as transplantation clinical periodontology.
