Identification of Pythium oligandrum using species-specific ITS rDNA PCR oligonucleotides

摘要: Pythium oligandrum is a parasite of cultivated Agaricus bisporus. Infection results in significant yield reductions and disease referred to as ‘black compost’. In this study, P. isolates were isolated from New Zealand mushroom composts, their ribosomal DNA internal transcribed spacer (ITS) regions amplified using the polymerase chain reaction (PCR). ITS nucleotide sequences obtained compared with those previously identified 23 described species. Although had high identity internationally oligandrum, order nucleotides some varied when other These within region used design PCR primers (P.OLIG.F1 P.OLIG.R04) for specific amplification 384-bp fragment DNA. P.OLIG.F1 P.OLIG.R04 identify major source inoculation on farm. Application diagnostic test will assist farming strategies implemented prevent future outbreaks.
