摘要: Tyrosine phosphorylation exerts a pivotal role in cell regulation processes of higher eukaryotes. Tight control the activity protein tyrosine kinases is crucial for ordered to occur. We have developed functional screen kinase regulators using c-Src, first cellular described, as prototype; and fission yeast, Schizosaccharomyces pombe, genetically amenable host system. Inducible expression c-Src yeast lethal. screened human cDNA libraries clones able counteract lethal effect Src. Two different classes cDNAs, which we called SAS sequences antagonizing Src, were obtained. The class encodes Csk, known regulate Src through C-terminal tyrosine. second consists encoding three phosphatases, counteracting action by dephosphorylation substrates itself. system described here can be applied identify other heterologous kinases, including receptor-type impair growth S. pombe.
