A Novel DNA-Binding Protein with Homology to Myb Oncoproteins Containing Only One Repeat Can Function as a Transcriptional Activator

摘要: A cDNA clone encoding a novel Myb-related protein, designated MybSt1, was isolated from potato expression library by South Western screening using the CaMV 35S promoter domain as probe. Sequence comparison shows small region with some homology to highly conserved DNA binding of c-myb proto-oncogene consisting three imperfect repeats. The Myb motif MybSt1 protein is distinct plant described so far. In contrast known proteins, two repeats required for activity, mybSt1 contains only one such repeat. Nevertheless, able bind in sequence-specific manner. addition Myb-like region, an acidic segment its central well proline-rich near C-terminus. Applying random site selection technique, high-affinity sites were identified, sharing core GGATA. transient assays protoplasts, clear evidence obtained this myb functioning transcriptional activator.
