摘要: The Hs1pro-1 gene confers resistance to the beet cyst nematode Heterodera schachtii in sugar (Beta vulgaris L.) on basis of a gene-for-gene relationship. RNA-gel blot analysis revealed that transcript was present uninfected roots resistant at low levels but increased by about fourfold one day after infection. Treatments plants with external stimuli including salicylic acid, jasmonic gibberellic acid and abscisic as well wounding or salt stress did not result changes transcription, indicating de novo transcription upon infection specifically. To study transcriptional regulation expression cellular level, 3082 bp genomic fragment representing promoter, isolated from YAC-DNA housing gene, fused beta-glucuronidase reporter (1832prm1::GUS) transformed into susceptible Arabidopsis plants, respectively. Fluorometric histochemical GUS assays transgenic carrying 1832prm1::GUS construct demonstrated promoter is functional both species drives responsive feeding site-specific GUS-expression. activity detected early initiation sites staining restricted sites. delineate regulatory domains fusion genes various 5' deletions were constructed analysed well. Cis elements responsible for reside between -355 +247 site whereas an enhancer region necessary higher located -1199 -705 promoter. suggesting conserved mechanism these two species.
