摘要: A novel pathway for the synthesis of poly-3-hydroxybutyrate has been engineered by simultaneous delivery two genes into insect cells (Spodoptera frugiperda) use individual baculovirus vectors. This system includes expression a dehydrase-domain mutant rat fatty acid synthase cDNA and phbC gene encoding polyhydroxyalkanoate from Alcaligenes eutrophus. The dehydrase-deficient provides de novo R-(-)-3-hydroxybutyryl-coenzyme as premature termination product rather than palmityl-coenzyme A, normal wild-type synthase. High levels this multifunctional protein provide suitable precursor pool conversion to in coexpressing product. strategy redesigning biosynthetic suggests new method generating structurally diverse polyhydroxyalkanoates metabolic engineering.
