摘要: Abstract Two species of double-stranded RNA (DS RNA) were isolated from TMV-infected leaf tissue with the aid cellulose chromatography. They have characteristics replicative form (RF) and intermediate (RI) found in other virus infected tissue. Their buoyant densities Cs 2 SO 4 are, respectively, 1.615 1.630 g/ml, whereas TMV-RNA has a density 1.640 g/ml. DS displays sharp thermal transition at 68° 10 −3 M potassium phosphate buffer containing Na EDTA is detected appreciable amount only cell fraction sedimenting mitochondria. RF RI estimated molecular weights, × 6 5.0 daltons as determined by polyacrylamide gel electrophoresis. not infectious but becomes so upon denaturation. It probably composed its complementary strand on basis above evidence because tritium-labeled partially resistant to nuclease action incubation denatured under annealing conditions.
