A novel micro-emulsion and micelle assembling method to prepare DEC205 monoclonal antibody coupled cationic nanoliposomes for simulating exosomes to target dendritic cells

摘要: Abstract Cationic biomimetic exosomes were prepared using a novel micro-emulsion and micelle assembling method by introducing DEC205 monoclonal antibody as specific ligand to target dendritic cells (DCs). The Box–Behnken experimental design was applied for optimization of nanoliposomes (NLip) then conjugated on the surface NLip (DEC205-NLip). DEC205-NLip respectively had an average size 62.7 ± 6.33 nm 81.64 ± 4.25 nm, zeta potential +30.5 ± 2.3 mV +19.8 ± 1.8 mV encapsulation efficiency 91.02 ± 3.1% 93.10 ± 2.2%. In addition, toxicity studies confirmed could significantly reduce cytotoxicity cationic lipid against DCs. And cellular uptake experiment evaluated significant targeting effect DC cells. conclusion, presented here prepare efficient approach develop antigen carriers DCs targeting.
