摘要: Tetraarsenic hexoxide (As4O6) has been used in Korean traditional medicine for the treatment of cancer since late 1980's, and arsenic trioxide (As2O3) is currently as a chemotherapeutic agent. Previous studies suggest that As4O6-induced cell death pathway different from As2O3 its mechanism anticancer activity remains unclear. Nuclear factor (NF)-κB well-known transcription involved proliferation, invasion metastasis. Hence, present study, we investigated effects As4O6 on NF-κB NF-κB-regulated gene expression vitro vivo. The cytotoxicity assay revealed inhibited growth SW620 cells dose-dependent manner, half maximal inhibitory concentration (IC50) was ~1 µM after 48 h treatment. suppressed activation κBα (IκBα) phosphorylation stimulated by tumor necrosis (TNF). also downstream proteins anti-apoptosis, In addition, marginally anti-NF-κB confirmed using an vivo xenograft mouse model which animals were injected with cells. study provides evidence properties through suppression NF-κB-mediated cellular responses.
