摘要: Abstract In this study, we constructed particles applicable for oral immunotherapy of type I allergy by protecting allergens from digestion and supporting intestinal antigen uptake. Therefore, birch-pollen were entrapped in poly( d , l -lactic-co-glycolic acid) microspheres spray-drying rendering with a main population 1–3 μm. Microspheres further coated wheat germ agglutinin (WGA) to target enterocytes. Coating WGA did not alter the surface characteristics as demonstrated scanning electron microscopy. Binding was specific could be inhibited chitotriose 14.7 ± 6.9%. Comparable amounts allergen released both particle-types 46.3 ± 1.7% 44.5 ± 2.6% during 21 days. Simulating gastric vitro, antigenicity WGA-microspheres preserved 59.8 ± 1.5% even after 2 h. Feedings BALB/c mice induced higher levels allergen-specific IgG-levels than gavages uncoated microparticles or naked protein. Thus, conclude that are suitable vehicles delivery mucosal targeting due lectin-mediated bioadhesion.
