摘要: This study investigated the potential use of stem cell-synthesized extracellular matrix (ECM) as a bone graft replacement therapy. ECM was synthesized by both mesenchymal cells (MSCs) and amniotic fluid (AFS cells) decellularized via freeze-thaw cycling DNase treatment. A series in vitro studies were performed to evaluate effect on cell attachment, proliferation, differentiation. Reseeded MSCs able attach proliferate types 2D 3D culture. In 2D, cultured showed increased levels calcium deposition. 3D, mineralized production reseeded MSCs, with produced AFS inducing greater amount formation than MSC ECM. Both used treat rat femoral segmental defect vivo. Each type rate bridging when compared collagen coated scaffolds. However, presence did not have significant volume within site this study.
