摘要: The biosynthetic and structural characteristics of the human thymocyte/T cell antigen defined by monoclonal antibody WT1 have been studied. identified a monomeric surface glycoprotein Mr = 40,000 ( gp40 ). Cross-absorption experiments two-dimensional gel analyses indicate that another antibody, 3A1, react with same structure. This was asymmetrically inserted into rough endoplasmic reticulum as transmembrane At this stage, polypeptide chain possessed two N-linked, "high-mannose" type glycans; these were subsequently processed endo-H-insensitive, complex oligosaccharides during intracellular transport to surface. Inhibition N-linked glycosylation tunicamycin failed block processing nonglycosylated 29,000 33,000. Cleavage mature form endo-F yielded similar 33,000 product. kinetics synthesis intermediate in conjunction gal-NAc oligosaccharidase digestion indicated presence O-linked glycans antigen. fully also contains covalently associated fatty acid, labeled 32P phosphate, predominantly phosphoamino acid being phosphoserine. We demonstrate biochemically reactivity cells from few patients acute myeloid leukemia reflects genuine expression structure on cells.
