摘要: Oxygen radical production by mitochondria seems to be implicated in various degenerative diseases and aging. In order estimate the rate of generation reactive oxygen species, it is necessary use adequate methods. this article we describe a fluorometric method appropriate measure H 2 O isolated functional mitochondria. The specific for sensitive enough assay mitochondrial presence respiratory substrates without inhibitors chain. Just after isolating from fresh tissues, rates are measured fluorometry homovanillic acid horseradish peroxidase. Simultaneous measurement consumption allows calculation percent electrons out sequence, which reduce radicals along chain instead reducing water at terminal cytochrome oxidase. This known as free leak. also study effect different modulators on production, allowing localization sites where species specifically formed.
