Characterization of functional endothelin receptors in the porcine myometrium.

摘要: To characterize the endothelin (ET) receptor that mediates contraction induced by ET-1 in porcine myometrium, we carried out a study, radioligand binding study and molecular (reverse transcription polymerase chain reaction) for detection of ET receptor-coding genes (mRNA). (1 nM-1 microM) caused tetrodotoxin-insensitive both longitudinal circular muscles, but muscle was more sensitive to than muscle. On other hand, ET-3 sarafotoxin S6c were less effective cause contractile response. The markedly inhibited BQ123 FR139317, BQ788 only slightly response ET-1. indicated presence single class 125I-ET-1 sites with same Kd value layers. However, Bmax (3252 fmol/mg protein) significantly higher (1883 protein). FR139317 specific completely, ET-3, BQ3020 (inhibition, 10-20%), suggesting ET(A) is dominant subtype myometrium. results expression ET(B) In conclusion, causes myometrium through activation present on smooth cells. There marked layer-related difference (longitudinal > muscle) ET-1-induced concentration.
