Microsatellite DNA somaclonal variation in micropropagated trembling aspen (Populus tremuloides)

摘要: Microsatellite DNA markers of ten simple sequence repeat (SSR) loci were used to examine somaclonal variation in randomly selected micropropagated plantlets derived from three different Populus tremuloides donor trees (genotypes). The obtained tissue cultures dormant vegetative buds, and those the same tree, grown greenhouse, did not exhibit any sign visible morphological variation. No microsatellite was observed among 13 somaclones one tree 4 another at eight SSR loci. However, despite small number progeny per sampled, detected two primer pair for locus PTR5 revealed 1 out genotype, while PTR2 four genotype. resulted appearance a new allele increased size, possibly due an addition units, third allele, presumably presence single extra chromosome or duplication chromosomal segment. These results demonstrate that genetic fidelity plants P. cannot always be assured can occur even when tissues well organized buds are cultures; gross level; provide useful sensitive determining clonal tremuloides.
