Innate and adaptive immunity gene expression of human keratinocytes cultured of severe burn injury

摘要: PURPOSE: Evaluate the expression profile of genes related to Innate and Adaptive Immune System (IAIS) human Primary Epidermal keratinocytes (hPEKP) patients with severe burns. METHODS: After obtaining viable fragments skin without burning, culture hKEP was initiated by enzymatic method using Dispase (Sigma-Aldrich). These cells were treated Trizol(r) (Life Technologies) for extraction total RNA. This quantified analyzed purity cDNA analysis gene specific IAIS PCR Arrays plates (SA Biosciences). RESULTS: we found that 63% these differentially expressed, which 77% repressed 23% hyper-regulated. Among these, following (fold increase or decrease): IL8 (41), IL6 (32), TNF (-92), HLA-E (-86), LYS (-74), CCR6 (- 73), CD86 (-41) HLA-A (-35). CONCLUSIONS: study contributes understanding molecular mechanisms underlying wound infection caused burn. Furthermore, it may provide new strategies restore normal thereby change healing process improve clinical outcome.
