Panel of monoclonal antibodies to sperm surface proteins as a tool for monitoring localization and identification of sperm–zona pellucida receptors

摘要: Primary binding of the sperm to zona pellucida (ZP) is one many steps necessary for successful fertilization. Sperm bind ZP by means membrane receptors which recognize carbohydrate moieties on glycoproteins according a well-defined sequential process. receptors, have been disclosed in various mammals, are localized throughout acrosomal region surface. A panel monoclonal antibodies against proteins from surface was prepared. Antibodies were screened immunofluorescence protein localization and Western blotting. Proteins head simultaneously detected blotting further studied terms immunolocalization reproductive tissues fluids, ZP, immunoprecipitation sequencing. Of 17 prepared antibodies, 8 recognized also interest Only three other that coincided ZP. These used immunoprecipitation, sequencing immunoprecipitates revealed these distinguished acrosin precursor, RAB-2A protein, lactadherin P47. This not first time we ejaculated capacitated sperm. However, our knowledge, this has Lactadherin P47 already characterized its physiological function reproduction proposed.
