摘要: The demonstration of immunoglobulin or T-cell receptor gene rearrangements in human lymphoproliferative processes with the use DNA hybridization has gained great popularity as a sensitive laboratory adjunct to diagnostic hematopathology. fact that nearly all B- malignant lymphomas and leukemias have one more rearranged antigen genes provides biologic basis for test. To formally analyze sensitivity, specificity, reproducibility diagnosis disease, authors conducted large, multiinstitutional study. Through blinded, controlled approach, rearrangement analysis 275 cases was shown carry high correlation conventional phenotyping histologic diagnosis, only minor false-positive false-negative rates. Significantly, no were detected normal lymphoid tissues carcinomas, sarcomas, melanomas. In randomized study 50 cases, results showed rate interlaboratory agreement, regardless level previous experience. Furthermore, interpretation data (Southern blot autoradiograms) 192 concordance among 11 observers from multiple laboratories.
