摘要: The classical small molecule neurotransmitters are essential for cell-cell signaling in the nervous system regulation of behaviors and physiological functions. Metabolomics approaches ideal quantitative analyses neurotransmitter profiles but have not yet been achieved repertoire 14 neurotransmitters. Therefore, this study developed targeted metabolomics by full scan gas chromatography/time-of-flight mass spectrometry (GC-TOF) hydrophilic interaction chromatography-QTRAP (HILIC-MS/MS) operated positive ionization mode identification quantitation consisting acetylcholine, adenosine, anandamide, aspartate, dopamine, epinephrine, GABA, glutamate, glycine, histamine, melatonin, norepinephrine, serine, serotonin. GC-TOF represents a new method analyses. Sensitive measurements 11 were GC-TOF, three analyzed LC-MS/MS (acetylcholine, melatonin). limits detection (LOD) (LOQ) assessed linearity protocols. In neurotransmitter-containing dense core secretory vesicles adrenal medulla, known as chromaffin granules (CG), measured concentrations 9 catecholamines combined with serotonin, serine. CG constitutively secreted from sympathoadrenal cells culture. Nicotine- KCl-stimulated release adenosine. Lithium, drug used treatment bipolar disorder, decreased constitutive secretion dopamine norepinephrine nicotine-stimulated epinephrine. Lithium had no effect on other Overall, newly methods will benefit investigations biological systems human disease conditions related to treatments.
