摘要: Benzene is a recognized hematotoxin and leukemogen, but its mechanisms of action in humans are still uncertain. To provide insight into these processes, we carried out cross-sectional study 44 healthy workers currently exposed to benzene (median 8-hr time-weighted average; 31 ppm), unexposed controls Shanghai, China. Here an overview the results on peripheral blood cells levels somatic cell mutation frequency measured by glycophorin A (GPA) gene loss assay report cytokine levels. All (i.e., total white cells, absolute lymphocyte count, platelets, red hemoglobin) were decreased among compared controls, with exception mean corpuscular volume, which was higher subjects. In contrast, (interleukin-3, interleukin-6, erythropoietin, granulocyte colony-stimulating factor, tissue necrosis factor-alpha) subset most highly (n = 11) similar values 11), suggesting that does not affect growth factor blood. The GPA measures stem or precursor erythroid mutations expressed MN heterozygous subjects, identifying NN variants, result from M allele duplication N allele, phi arise inactivation. (but phi) variant elevated (mean +/- SD, 13.9 8.4 mutants per million versus 7.4 5.2 (respectively; p 0.0002), produces gene-duplicating gene-inactivating at locus bone marrow humans. These findings, combined ongoing analyses macromolecular adducts chromosomal aberrations, will opportunity comprehensively evaluate wide range early biologic effects associated exposure
