摘要: Intervertebral disc (IVD) degeneration (IDD) is the most common cause leading to low back pain (LBP), which a highly prevalent, costly, and crippling condition worldwide. Current treatments for IDD are limited treat symptoms do not target pathophysiology. Tumor necrosis factor-α (TNF-α) one of potent pro-inflammatory cytokines signals through its receptors TNFR1 TNFR2. TNF-α expressed in degenerative IVD tissues, it deeply involved multiple pathological processes degeneration, including matrix destruction, inflammatory responses, apoptosis, autophagy, cell proliferation. Importantly, anti-TNF-α therapy has shown promise mitigating relieving LBP. In this review, following brief description signal transduction, we mainly focus on expression pattern roles IDD, summarize emerging progress regarding inhibition as promising biological therapeutic approach associated A better understanding will help develop novel TNF-α-centered interventions disease.
