摘要: Background: Growth-differentiation factor 15 (GDF15) is a member of the transforming growth β (TGF-β) cytokine superfamily. There has been increasing interest in using circulating GDF15 as biomarker patients, for example those with cardiovascular disease. Methods: We developed an IRMA that uses polyclonal, affinity chromatography–purified goat antihuman IgG antibody, assessed preanalytic characteristics GDF15, and determined concentrations 429 apparently healthy elderly individuals 153 patients chronic heart failure (CHF). Results: The assay had detection limit 20 ng/L, intraassay imprecision ≤10.6%, interassay ≤12.2%. Specificity was demonstrated size-exclusion chromatography, parallel measurements polyclonal monoclonal anti-GDF15 lack cross-reactivity TGF-β. not appreciably influenced by anticoagulant matrix or unrelated biological substances. stable at room temperature 48 h resistant to 4 freeze-thaw cycles. Apparently healthy, presented median concentration 762 ng/L (25th–75th percentiles, 600–959 ng/L). were associated age cystatin C C-reactive protein concentrations. CHF increased closely related disease severity. Conclusion: can detect human serum plasma excellent sensitivity specificity. reference limits confounding variables defined favorable are expected facilitate future studies various settings, including CHF.
