摘要: The imprinting of mammalian genes depends on the maintenance DNA methylation patterns during pre- and postimplantation development. Dnmt1o is a variant form somatically expressed Dnmt1 cytosine methyltransferase that synthesized stored in oocyte cytoplasm trafficks to eight-cell nucleus preimplantation development, where it maintains alleles imprinted genes. Transcripts encoding are present embryos, suggesting protein also embryo, may account for at stages other than embryo. However, using an antibody detects Dnmt1, but not Dnmt1o, no was detected immunoblots or by immunocytochemical staining wildtype embryos. Moreover, produced from modified allele, Dnmt1(1s/1o), trafficked nuclei stages. highly restricted nuclear localization oocyte-derived development add further support notion methyltransferases required maintaining imprints
