摘要: Cold pretreatment, washing medium and composition of nutrient media may have marked effects on microspore embryogenesis. When microspores isolated from radish (Raphanus sativus L. cv. Gwanhun) flower buds were washed with Nitsch & (NLN) liquid containing 130 g?L -1 sucrose (NLN-13), yields microspore-derived embryos greater than when using B5 sucrose. Microspore viability is known to decrease rapidly storage; however, in this experiment, was maintained for 24 h at 4°C without media. Among the various concentrations used (0.25×, 0.5×, 1.0×, 2.0×, 4.0× NLN medium), 0.5× induced most efficient formation embryos. In addition, cultured supplemented 0.25×, 1.0× microelements, compared not microelements. study, highest yield observed derived NLN-13 then 0.25× followed by incubation 25°C 30 days.
