Lipids that determine detergent resistance of MDCK cell membrane fractions.

摘要: A comparative lipidomic study has been performed of whole Madin-Darby canine kidney epithelial cells and the detergent-resistant membrane fraction (DRM) obtained after treating with non-ionic detergent Triton X-100. The DRM were isolated following a standard procedure that is extensively used in cell biology studies. Significant differences found lipid composition DRM. latter enriched all analyzed sphingolipid classes: sphingomyelins, ceramides hexosylceramides. Diacylglycerols also preferentially was saturated over unsaturated fatty acyl chains, sn-1 chains containing 16 carbon atoms, longer shorter ones. glycerophospholipid species phosphatidylethanolamines phosphatidylinositols, mainly unsaturated, did not show preference for Phosphatidylcholines an intermediate case: saturated, but question remains on whether these DRM, recovered from detergent-membrane mixtures by floatation sucrose gradient, really correspond to domains existing prior treatment.
