摘要: Lomandra sonderi (F.Muell.) Ewart (Asparagaceae) is endemic to the south-west Western Australian jarrah (Eucalyptus marginata Donn ex Sm.) forest region, and a difficult propagate species important post-mining restoration. Micropropagation only way currently produce plants of this for This study describes investigations into optimising cryopreservation efficient long-term germplasm storage. In order investigate effect preconditioning on post-cryogenic survival shoot tips, in vitro grown were exposed range light-, temperature- osmotic-induced treatments under culture room conditions 3 weeks. Room temperature (24°C) resulted greatest survival, followed by low light (17 µmol m–2 s–1) preconditioning. Alternating (25/5°C), high (35°C), sucrose (180 mM) (93 µmol m–2 s–1) all led significantly progressively lower tip than Antioxidant activity superoxide dismutase preconditioned tips showed positive correlation overall, whereas activities glutathione reductase, peroxidase catalase little correlation. Analysis throughout protocol that reductase decreased after cryopreservation, whilst did not change.
