摘要: Ribosome display is an in vitro evolution technology for proteins. It based on translation, but prevents the newly synthesized protein and mRNA encoding it from leaving ribosome. thereby couples phenotype genotype. Since no cells need to be transformed, very large libraries can used directly selections, amplification provides a convenient integration of random mutagenesis that incorporated into procedure. This review highlights concepts, mechanisms, different variations ribosome compares related methods. Applications are summarized, e.g., directed proteins higher binding affinity, stability or other improved biophysical parameters enzymatic properties. has developed robust academia industry alike, made cell-free Darwinian over multiple generations reality.
