摘要: Publisher Summary The polysome display system is a recent biological method for screening peptide libraries. Unlike the cell-based methods, relies on in vitro synthesis and amplification of sequences during each round screening. This offers potential larger libraries more diverse collection since few biases should affect gene expression . A notable difference between phage current affinities recovered peptides. It found that high-affinity (Kd values less than 200 nM) predominate pools enriched suggests may be monovalent. contrast to multivalent plasmid systems can recover ligands, with as low 100μM. Monovalent limits range ligand families are recovered, but very effective selecting higher affinity sequences, especially if mutagenic PCR 22 incorporated rounds selection.
