摘要: Publisher Summary This chapter discusses the in vitro translation, which is a prerequisite for devising and optimizing cell-free protein selection systems. In synthesis, independent of its use technology, this due to improvement yields, increase detection sensitivity many analytical methods, advent new technologies, such as atomic force microscopy (AFM) fluorescence correlation spectroscopy (FCS) that allow work at low concentrations, even down single molecule level. Many applications translation rely on correct folding expressed polypeptide into three-dimensional structure, all systems are based translation. Because proteins selected evolved functionality, sufficient expression respective system necessity efficient selection. An attractive advantage using translations that, least principle, any component reaction can be deliberately added or removed. To achieve yield, however, separate consideration both actual necessary. Even optimized systems, yields not similar globular proteins.
