摘要: Receptor-mediated endocytosis involves the transport of receptor-ligand complexes from cell surface to an intracellular endocytic compartment. This study shows that plasma membrane-derived vesicles containing receptor-bound ligands (e.g. aggregated anti-dinitrophenol (DNP) IgG bound Fc receptors) fuse with early endosomes DNP-beta-glucuronidase in a cell-free system. Plasma membrane were generated by homogenization cells had been allowed bind at 4 degrees C. Fusion between two probes was assessed (i) formation anti-DNP IgG-DNP-beta-glucuronidase and (ii) colocalization within closed different sizes colloidal gold coated ligands. required ATP, cytosol, KCl. The requirements similar those described for endosome-endosome fusion vitro systems. Mild trypsinization prior their addition assay inhibited fusion. When chased into more mature compartments, not observed. results indicate regions involved receptor-mediated are capable fusing endosomes. event may constitute first step
