摘要: Human airway smooth muscle cells (HASMC) contribute to the process of wall remodelling in asthma by virtue their secretory functions. This study was performed investigate effectiveness commonly used steroids beclomethasone, budesonide and fluticasone downregulating HASMC production RANTES IL-8. (n=5) were cultured from dissected bronchi using collagenase digestion. Confluent exposed TNFalpha IL-1beta (10 ng/ml) for 24 h. All stimulations set with without pre-treatment or 2 h at concentrations 10(-9)-10(-6)M. IL-8 mRNA expression assessed RT-PCR protein secretion determined ELISA. Pre-treatment reduced TNFalpha- IL-1beta-stimulated release a dose dependent manner. However, beclomethasone 22-28% less effective than inhibiting chemokine production. IL-1beta-induced on transcriptional level budesonide. The results suggest that topical fluticasone, lesser extent may have beneficial effects inflammation reducing IL-8-induced leukocyte infiltration into wall.
