摘要: A novel protein sample pretreatment method based on ampholine immobilized polymer microsphere (ampholine@PM) was developed for the fractionation of intact proteins prior to digestion and peptide analysis reduce dynamic range human plasma proteome. After incubation with our prepared ampholine@PM, captured were successively desorbed by 2 M NaCl, 100 mM glycine-hydrochloric acid, 30% (v/v) acetonitrile 0.1% trifluoroacetic acid. The SDS-PAGE results showed in such three fractions obviously reduced as compared native plasma. On-particle ultimately performed release all retained ampholine@PM. Followed MuPIT analysis, number identified improved 75% after ampholine@PM treatment. Furthermore, spectral count 9 high abundance decreased 37.6-97.2%, low (< ng mL(-1)) increased from 4 17.
