摘要: Effects on translation in vivo by modification deficiencies for 2-methylthio-N6-isopentenyladenosine (ms2i6A) (Escherichia coli) or 2-methylthio-N6-(4-hydroxyisopentenyl)adenosine (ms2io6A) (Salmonella typhimurium) tRNA were studied mutant strains. These hypermodified nucleosides are present the 3' side of anticodon (position 37) reading codons starting with uridine. In E. coli, translational error caused was strongly reduced case third-position misreading a tryptophan codon (UGG) particular context but not affected first-position an arginine (CGU) another context. Misreading UGA nonsense at two different positions dependent. The efficiencies some suppressors decreased tRNA-dependent manner. Suppressor which lacks ms2i6A-ms2io6A becomes more sensitive to Our results therefore indicate that, besides improving efficiency, ms2i6A37 and ms2io6A37 modifications also involved decreasing intrinsic sensitivity tRNA. Possible consequences regulation gene expression discussed.
