摘要: Abstract The effect of taurocholate on transcytotic vesicular pathways labeled with horseradish peroxidase was assessed in isolated perfused rat liver preparations. Forty-five minutes after a load recirculating system, continuous infusion but not taurodehydrocholate significantly increased excretion bile by 50% compared controls. When (25 mg) pulse loaded for 1 minute control perfusions, it appeared early (4–6 minutes) and late (20–25 peaks, the latter accounting 90% total output. Taurocholate output both whereas only small increase peak observed taurodehydrocholate. Colchicine pretreatment abolished second peak. Electron micrographs from livers revealed accumulation peroxidase-containing vesicles pericanalicular regions at (2 as well (18 periods. morphometric analysis electron performed 2 1-minute (500 mg), density percent area In contrast, colchicine had no early-appearing vesicles, although their individual sizes were reduced. also tubular structures region. These findings indicate that stimulates vesicle rapid transcellular-colchicine-insensitive therefore probably microtubule-independent pathway is present hepatocytes must be distinguished paracellular routes.
