Development of multilocus sequence typing (MLST) assay for Mycoplasma iowae.

摘要: Abstract Mycoplasma iowae (MI) infection is an economically and commercially important disease of turkeys. There are no sequence typing assays available for MI strain identification, the only molecular tools this purpose, DNA fingerprinting assays. In addition to their low reproducibility, require isolation microorganism in pure culture, which difficult avian mycoplasma. Therefore, we propose a multilocus (MLST) assay as first genotyping identification MI. Based on two genomes GenBank, 26 loci housekeeping genes were identified studied diverse sample set. Finally, six selected newly developed MLST assay. The final analysis (total 5019 bp) (dppC, ulaA, valS, rpoC, leuS, kdpA) allowed differentiation 47 samples into 23 unique types. Moreover, when 4 used type same set samples, they resulted 20 Analysis phylogenetic trees clonal groups generated by displayed high degree agreement with geographical temporal information tested samples. highly reproducible epidemiology that can be identify positive clinical cases directly from it provides useful tool allowing better control eradication efforts.
