摘要: The present invention describes methods to construct circularization indexers and use them selectively circularize, purify, manipulate nucleic acids from a complex acid mixture. An assay is provided detect without any amplification. Kits for preparing indexing targets high-level multiplexing DNA amplification are provided. also method assembly, in which the self-primed need external primers. In this method, double stranded oligonucleotides with two cohesive ends (C-indexers) used circularize double-stranded or single-stranded such way that one of strands completely circularized while other strand either contains nick (if double-stranded) only hybridized part single-stranded). At presence polymerase, un-circularized-strand can serve as primer amplify primer.
