摘要: High-performance liquid chromatography (HPLC) was combined on-line with electrospray ionization mass spectrometry (ESI-MS) for structural analysis of a synthetic osteocalcin derivative and its degradation products. Initial determination amino acid sequence the peptide performed after tryptic degradation. Hydrolytic studied under different pH conditions: 2, 7 10 at 60°C up to 20 h. According HPLC-ESI-MS results, chemical stability dependent on pH. Two major products number other fragments were obtained in acidic solution, whereas molecule rather stable neutral alkaline conditions.
