A novel snake venom disintegrin that inhibits human ovarian cancer dissemination and angiogenesis in an orthotopic nude mouse model.

摘要: OVCAR-5 is a human epithelial carcinoma cell line of the ovary, established from ascitic fluid patient with progressive ovarian adenocarcinoma without prior cytotoxic treatment. The unique growth pattern makes it an ideal model for examining anticancer activity contortrostatin (CN), homodimeric disintegrin southern copperhead venom. FACS analysis revealed that integrin αvβ3 negative, but αvβ5 positive. CN effectively blocks adhesion cells to several extracellular matrix proteins and inhibits tumor invasion through artificial basement membrane. In xenograft nude mouse intraperitoneal introduction cells, injection was used therapy. Tumor dissemination in CN-treated versus control groups studied by gross examination, antiangiogenic potential examined factor VIII immunohistochemistry image analysis. not only significantly inhibited cancer model, also dramatically prevented recruitment blood vessels tumors at secondary sites.
