摘要: We have investigated the in vivo functional role of rab5, a small GTPase associated with plasma membrane and early endosomes. Wild-type rab5 or rab5-ile133, mutant protein defective GTP binding, was overexpressed baby hamster kidney cells. In cells expressing rab5ile 133 protein, rate endocytosis decreased by 50% compared normal, while recycling not significantly affected. The morphology endosomes also drastically changed which induced accumulation tubules vesicles at periphery cell. Surprisingly, overexpression wild-type accelerated uptake endocytic markers led to appearance atypically large conclude that is rate-limiting component machinery regulating kinetics traffic pathway.
