Estimation of genotyping errors of STR markers in dogs and wolves

摘要: Abstract This article will discuss the estimation of genotyping errors for Mini-DogFiler panel A and B in group wolfs (n = 39) where source material DNA isolation were faeces control dogs (n = 170), represented by Czechoslovakian wolfdog breed (CSW), was obtained from buccal swabs. samples isolated swabs had a very low frequency error estimated as mean rate per allele (ea = 0.09 %), locus (el = 0.18 %) multilocus genotype (eobs = 2.17 %). Completely different situation observed affirms how complicated is to get reliable results such material. Total performance poor only 7 out 39 replicated (∼18 at least 2 times 4 replicates. In case would be eobs = 86.5 %. For this type typical fragmented with concentration. fact leads occurrence many negative effects like an allelic dropout, null or presence PCR artefacts. Results confirm necessity multiple isolates one sample amplification these creating consensus each genotype. Moreover, both types suffered relatively high dropout occurring VGL2136, but it did not increase because no signal detected all cases. The cause behaviour remained unknown raised question whether appropriate analysis wolf CSW samples.
