Characterization of voltage-sensitive sodium channel gene coding sequences from insecticide-susceptible and knockdown-resistant house fly strains

摘要: The kdr insecticide resistance trait of the house fly (Musca domestica L.), which confers reduced neuronal sensitivity to DDT and pyrethroid insecticides, was previously shown exhibit tight genetic linkage restriction fragment length polymorphism markers lying within a voltage-sensitive sodium channel gene that is homologous para Drosophila melanogaster. In present study, 6315 nucleotide coding sequences this from insecticide-susceptible (NAIDM strain) (538ge flies were determined by automated direct DNA sequencing PCR fragments obtained amplification on first strand cDNA adult heads. deduced 2105-residue amino acid sequence each strain exhibited overall structure organization typical α subunit genes 90.0% identical D. melanogaster product. We did not detect any splice variants among cDNAs head preparations. Comparison NAIDM revealed 12 differences in 538ge strain. significance these polymorphisms as candidate resistance-conferring mutations discussed.
