摘要: Neutrophils (PMN) preincubated with recombinant human granulocyte/macrophage colony-stimulating factor (rhGM-CSF) for 2 h and then stimulated the chemotactic factors, C5a or FMLP, produce substantial amounts of lipoxygenase products 5-Hete, LTB4, omega-oxidised LTB4 metabolites (4.36 +/- 0.95 (SEM) pM (n = 21) metabolites/10(6) PMN). No are detected by HPLC RIA if purified PMN either GM-CSF factors in absence exogenous arachidonate. The priming effect upon induced generation lipid mediators is a relatively slow process, clearly evident after 1 optimal h. Leukotriene measurable 0.8 U GM-CSF/10(6) maximal 80 (10(-11)-10(-9) M). Upon activation primed leukotriene synthesis very rapidly. Already 2.5 min major present 20-OH 20-COOH LTB4. Our study shows that from endogeneous AA can be initiated through receptor mediated processes appropriately timed combination biological soluble inflammatory mediator peptides. Furthermore, these results indicate not only enhances effector cell functions but qualitatively change profile formed second triggering signal. Such mechanism might important amplifying responses. Alternatively, also have an intracellular autocoid role responsible enhancement other like oxygen radical release.
