摘要: The Aurora (Ipl1)-related kinases are universal regulators of mitosis. We now show that Aurora-A, in addition to Aurora-B, regulates kinetochore function human cells. A two-hybrid screen identified the component CENP-A as a protein interacts with Aurora-A. Aurora-A phosphorylated vitro on Ser-7, residue also known be targeted by Aurora-B. Depletion or Aurora-B RNA interference revealed is initially prophase manner dependent and this reaction appears required for subsequent Aurora-B-dependent phosphorylation well restriction inner centromere prometaphase. Prevention led chromosome misalignment during mitosis result defect attachment microtubules. Our observations suggest Ser-7 essential function.
