Effect of local secondary structure of mRNA on expression of recombinant human lymphotoxin in Escherichia coli

摘要: We substituted a few alternative codons close to the N-terminal coding region of human lymphotoxin (LT) gene for natural ones avoid stable local secondary structures mRNA, predicted in 5′-terminal 130 nucleotides mRNA. With this modification, we have overexpressed recombinant (rLT) inEscherichia coli under control trc promoter. Most produced rLT was soluble and active form.
