A monoclonal antibody shows discrete cellular and subcellular localizations of mGluR1α metabotropic glutamate receptors

摘要: The metabotropic glutamate receptor, mGluR1α, is postsynaptic in excitatory synapses many populations of neurons and mediates long-term responses. present study defines the distribution this receptor using a new, highly specific monoclonal antibody to mGluR1α. Overall immunostaining was similar that described previously with polyclonal antibodies, including prominent staining olfactory bulb, interneurons CA1 hippocampus stratum oriens/alveus, globus pallidus, thalamus, Purkinje cells outer dorsal cochlear nucleus little or low principal cerebral cortex hippocampus. Interestingly, well-known association mGluR1α receptors neocortical even more prevalent than noted antibodies. Ultrastructural studies cerebellum showed dense immunoperoxidase membranes densities perisynaptic extrasynaptic membranes, as well substantial cytoplasmic associated organelles, especially endoplasmic reticulum.
