Nucleotide specificity of the steroidogenic response of rat adrenal cell suspensions prepared by collagenase digestion.

摘要: A previously described method for preparing rat adrenal cell suspensions by use of collagenase has been modified. The sensitivity the preparation to stimulation ACTH and 3′,5′-cyclic AMP was investigated. dibutyryl derivative 30 times more effective, at concentrations tested, than parent compound. Concerning nucleoside/nucleotide specificity suspensions, 42 purine pyrimidine compounds IMP active, about 50% activity AMP. All 3′,5′- cyclic nucleotides tested showed synergism in presence 5.0 mM theophylline; most striking effect seen with GMP. 2′,3′- Cyclic GMP 2′,3′-cyclic UMP were potent inhibitors AMP-stimulated steroidogenesis preparations suspensions.
